In this project, we systematically quantified interactions between 148 representative, ligand-activated GPCRs and Gα subunits C-termini (GPCR data). For each receptor, we probed chimeric Gα subunit activation via a transforming growth factor-α (TGFα) shedding response in HEK293 cells lacking Gq/11 and G12/13 subfamilies, complemented with a NanoBiT-based G-protein dissociation assay.

Interrogation of the dataset identified known and novel GPCR coupling specificity features, including residues at the G-protein interface and intra-cellular loops, with a majority involved in a contact network likely mediating receptor activation. Using these features we built an ML based coupling predictor (PRECOG).
